The effect of diazepam on hypercapnia-induced changes in the intracellular Ca2+ concentration was investigated in the rat hippocampal slice preparation loaded with the Ca2+-sensitive fluorescent dye fura-2. Hypercapnia produced a significant rise in the intracellular Ca2+ level ([Ca2+]i), which was accompanied by a large increase in the resting intracellular K+ concentration ([K+]i). Diazepam (0.5-5 microM) produced a dose-related inhibition of the increase in [Ca2+]i. The effect of diazepam was not affected by the addition of 5 microM flumazenil, a specific benzodiazepine receptor antagonist, or by the addition of 10 microM naloxone. In slices pretreated with tetrodotoxin, a voltage-dependent Na+ channel blocker, and a Ca2+ channel blocker, diazepam did not affect the rise in [Ca2+]i induced by hypercapnia. These results suggest that the inhibitory effect of diazepam on hypercapnia-induced increase in [Ca2+]i is not mediated via the benzodiazepine receptors in the hippocampal slice preparation.8
